mov003-Fc-GrB is a novel granzyme B-based fusion protein designed to target Folate Receptor alpha (FRα). It consists of a single-chain variable fragment (scFv) derived from the mov003 antibody, fused to an engineered human IgG Fc dimerization domain and the cytotoxic human serine protease granzyme B (GrB). Upon binding to FRα, which is overexpressed in various solid tumors (such as ovarian and breast cancer) and liquid tumors (like acute myeloid leukemia), the construct is internalized. Once inside the cell, GrB is delivered to the cytosol where it triggers apoptosis. This targeted approach aims to minimize systemic toxicity associated with traditional therapies. Preclinical studies have shown potent cytotoxic activity in FRα-positive cell lines, including those resistant to the Bcl-2 inhibitor venetoclax.
