Overview

MOXR0916 + atezolizumab is a **combination therapy** consisting of MOXR0916, a humanized IgG1 monoclonal antibody that acts as an effector-competent agonist targeting OX40 (a T-cell costimulatory receptor), and atezolizumab, an FDA-approved monoclonal antibody checkpoint inhibitor targeting PD-L1. MOXR0916 stimulates effector T cells and reduces regulatory T cells through OX40 agonist activity, potentially enhancing antitumor immune responses. Atezolizumab blocks PD-L1 interaction, restoring and augmenting T-cell mediated antitumor immunity. The combination is being studied for synergistic immunomodulatory and antitumor effects in advanced solid tumors, with highest activity seen in settings such as urothelial carcinoma, renal cell carcinoma, and other advanced or metastatic solid malignancies. MOXR0916 is investigational, while atezolizumab is approved in several indications; their combined use is experimental and not yet approved for any indication[1][2][3][4][7].
