MP-101 is an investigational, low-dose oral formulation of mefloquine, specifically the (+)-enantiomer, developed by Mediti Pharma for the treatment of dementia-related psychosis (DRP), agitation, and aggression. Mefloquine, originally an antimalarial agent, acts as a potent inhibitor of gap junctions, with a high affinity for Connexin 36 (Cx36). By blocking these gap junctions, MP-101 is intended to modulate neuronal synchronization and connectivity, which are hypothesized to be dysregulated in various forms of dementia, including Alzheimer's disease, Parkinson's disease dementia, and dementia with Lewy bodies. The drug was evaluated in a Phase 2 clinical trial (NCT03044249) targeting neuropsychiatric symptoms across the dementia spectrum, but the study was subsequently terminated.
