MP0274 is a novel DARPin (Designed Ankyrin Repeat Protein) therapeutic candidate developed for the treatment of HER2-positive cancers. Unlike established anti-HER2 antibody drugs, MP0274 binds to two distinct non-overlapping epitopes on the HER2 receptor, inhibiting its activity and promoting internalization. This action blocks HER2-mediated signaling pathways, induces apoptosis, and inhibits the growth of tumor cells that overexpress HER2. Preclinical studies have shown that MP0274 can also inhibit downstream signaling mediated by other members of the human epidermal growth factor receptor family (HER1/EGFR and HER3), providing broad anti-HER activity. The drug has demonstrated sustained tumor regression in clinical settings for patients with metastatic breast cancer who have failed prior lines of therapy targeting HER2. Its mechanism involves stabilization of AKT-regulated transcription factors leading to apoptosis, which may be unique among current anti-HER2 therapies[1][3][6][7].
