MP0317 is a tri-specific designed ankyrin repeat protein (DARPin)-based agent that targets the tumor-associated fibroblast activation protein (FAP), the CD40 receptor, and human serum albumin (HSA). It acts as a CD40 agonist that is exclusively activated in the presence of FAP expressed by cancer-associated fibroblasts within the tumor microenvironment. This design enables localized activation of immune cells—such as dendritic cells and B cells—within tumors while minimizing systemic toxicity. The drug aims to remodel the tumor microenvironment by increasing antigen-presenting cell abundance and enhancing dendritic cell maturation and IFNγ production. Developed for advanced solid tumors, it has demonstrated favorable safety in Phase 1 trials with evidence of pharmacodynamic activity at doses ≥1.5 mg/kg[1][2][3][4][6].
