MP0533 is a novel tetra-specific T cell-engaging Designed Ankyrin Repeat Protein (DARPin) therapeutic developed for the treatment of acute myeloid leukemia (AML) and myelodysplastic syndrome (MDS)/AML. It simultaneously targets three tumor-associated antigens—CD33, CD123, and CD70—on AML cells as well as the immune activator CD3 on T cells. This design enables avidity-driven binding that preferentially directs cytotoxic T cell activity against malignant cells co-expressing at least two of these antigens while sparing healthy cells that express only one or none. The mechanism results in selective killing of AML blasts and leukemic stem cells with reduced off-tumor toxicity compared to single-target agents. MP0533 has demonstrated tumor-localized T-cell activation, effective tumor regression in preclinical models, and lower cytokine release than other single-antigen targeting engagers. It is currently being evaluated in a first-in-human phase 1/2a clinical trial for relapsed/refractory AML or MDS/AML[1][4][6][7].
