Overview

MP0533 + obinutuzumab is a combination therapy being developed by Molecular Partners AG for the treatment of acute myeloid leukemia (AML) and myelodysplastic syndrome (MDS). MP0533 is a novel tetra-specific T-cell engaging DARPin (Designed Ankyrin Repeat Protein) that simultaneously targets three tumor-associated antigens—CD33, CD123, and CD70—on AML cells, while also binding to CD3 on T cells to induce tumor cell killing. Obinutuzumab, an anti-CD20 monoclonal antibody, is administered as a pretreatment in clinical protocols to mitigate the loss of drug exposure. This combination is currently being evaluated in a Phase 1/2a clinical trial (NCT05673057) to assess safety, tolerability, and preliminary efficacy in patients with relapsed/refractory or treatment-naïve AML and MDS.
