MP0621 is a multispecific, logic-gated protein therapeutic from Molecular Partners’ Switch-DARPin platform. It is designed as a next-generation conditioning agent for hematopoietic stem cell transplantation (HSCT), with the goal of selectively eradicating hematopoietic stem cells (HSCs) while minimizing toxicity associated with traditional high-intensity regimens. MP0621 targets three proteins: cKit (critical for HSC maintenance and renewal), CD16a (to engage innate immune cells such as natural killer cells and macrophages), and conditionally blocks CD47 (“don’t-eat-me” signal) only on targeted cKit-positive cells. This conditional blockade enhances the killing of HSCs by immune effector cells while reducing off-target effects seen with systemic anti-CD47 therapies. The drug’s logic-gated “Switch” mechanism ensures that immune activation occurs only in the presence of defined antigens, providing reversible and highly selective activity. As of mid-2024, MP0621 is in preclinical/IND-enabling studies, with Phase 1 trials in acute myeloid leukemia (AML) anticipated to begin in 2025[1][3][5][6].
