MP0712 is a first-in-class radiopharmaceutical drug candidate co-developed by Molecular Partners and Orano Med. It is a Radio-DARPin Therapeutic (RDT) that targets delta-like ligand 3 (DLL3), a protein highly expressed in small cell lung cancer (SCLC) and other aggressive neuroendocrine tumors but minimally present in healthy tissues. The drug consists of a DARPin protein engineered for high affinity and specificity to DLL3, conjugated with the alpha particle-emitting radioisotope lead-212 (\(^{212}\)Pb). This design enables targeted delivery of potent alpha radiation to DLL3-expressing tumor cells while minimizing off-target toxicity, particularly to the kidneys. Preclinical studies have demonstrated strong tumor uptake, favorable safety profile, and significant antitumor efficacy at clinically relevant doses. Clinical development is expected to begin in 2025 with initial data anticipated later that year[1][4][5][6][7].
