MP1032 is a first-in-class, orally administered small molecule drug developed by MetrioPharm for the treatment of immune-inflammatory and infectious diseases. It acts as a self-regulating modulator of mitochondrial metabolism in macrophages, targeting pathologically elevated reactive oxygen species (ROS) at sites of inflammation without affecting physiological ROS levels essential for normal cell function. Its mechanism involves scavenging ROS only when levels are abnormally high—activation is triggered by clusters of elevated pH in inflamed tissues. MP1032 downregulates pro-inflammatory cytokines such as TNFα, IL-1β, and IL-6 via normalization of cellular redox balance and modulation of Poly(ADP-ribose) polymerase 1 (PARP-1) activity. The drug has demonstrated anti-inflammatory effects comparable to corticosteroids but with an improved safety profile due to its targeted action. In addition to its anti-inflammatory properties, MP1032 has shown host-mediated antiviral and antibacterial activities in preclinical models and clinical trials have been conducted for COVID-19 infections, plaque psoriasis, Duchenne muscular dystrophy (with orphan drug status), multiple sclerosis, rheumatoid arthritis, juvenile rheumatoid arthritis, inflammatory bowel diseases, and other indications[1][3][4][5][6][7][8].
