MP71-MC2-TCR16-3D9 is an autologous T-cell receptor (TCR) engineered T-cell therapy developed by Erasmus University Medical Center (Erasmus MC). The therapy utilizes the MP71 gamma-retroviral vector to transduce a patient's own T cells with a TCR (clone 16-3D9) specific for the MAGE-C2 (Melanoma-associated antigen C2) protein. MAGE-C2 is a cancer-germline antigen that is highly expressed in several cancers, including melanoma and head and neck squamous cell carcinoma, but has limited expression in normal tissues, making it a suitable target for immunotherapy. The TCR recognizes the MAGE-C2-derived peptide ALKDVEERV presented by HLA-A*02:01. This cell therapy is designed to selectively identify and eliminate MAGE-C2-expressing tumor cells through direct cytotoxic activity upon recognition of the peptide-MHC complex.
