MPC-8640 is a small molecule prodrug that was developed by Myrexis (formerly Myriad Pharmaceuticals) for the treatment of various cancers. It functions by targeting cancer metabolism, specifically acting as a prodrug of an inhibitor of nicotinamide phosphoribosyltransferase (NAMPT). NAMPT is the rate-limiting enzyme in the NAD+ salvage pathway, which is frequently upregulated in cancer cells to meet high metabolic demands and support DNA repair. By inhibiting NAMPT, MPC-8640 aims to deplete intracellular NAD+ levels, leading to metabolic collapse and apoptosis in tumor cells. The compound was in the IND-enabling phase of development before Myrexis discontinued its internal research and development activities and liquidated its assets in 2012.
