mPEG112-b-PBLG5 (HIP) is an experimental nanotherapeutic consisting of a HuR-inhibiting peptide (HIP) formulated with a methoxy poly(ethylene glycol)-block-poly(gamma-benzyl-L-glutamate) (mPEG-b-PBLG) block copolymer. The drug is designed to target Human Antigen R (HuR), also known as ELAV-like protein 1, which is an RNA-binding protein that stabilizes the mRNA of pro-inflammatory cytokines. By inhibiting the structural interaction between HuR and its target RNAs, the HIP peptide prevents the downstream expression of inflammatory mediators. The mPEG112-b-PBLG5 copolymer serves as a delivery vehicle, forming polymeric micelles that enhance the stability and targeted delivery of the peptide. This therapeutic strategy is being investigated for the treatment of immune-mediated inflammatory diseases, specifically psoriasis, where it has demonstrated efficacy in reducing skin inflammation in preclinical mouse models.
