Overview

MPER-656 is a peptide-liposome therapeutic vaccine candidate designed to elicit immune responses against the membrane-proximal external region (MPER) of the HIV-1 envelope glycoprotein gp41. The vaccine consists of an MPER peptide (amino acids 656–680 of gp41) formulated in liposomes and adjuvanted with alum, administered intramuscularly. Its mechanism involves inducing antibodies and T-cell responses targeting the conserved gp41 region, which is recognized by broadly neutralizing antibodies (bnAbs). In a phase 1 clinical trial (HVTN 133), MPER-656 demonstrated robust immunogenicity, eliciting both antibody and CD4+ T-cell responses in most recipients. However, the study was halted early due to an anaphylactic reaction attributed to polyethylene glycol in the formulation. The product is being developed primarily for prevention or treatment of HIV infection by stimulating neutralizing immune responses[1][3][4][7].
