**mPF gene therapy** is an investigational viral vector-delivered gene therapy for glioblastoma that introduces a murine transcription-factor cassette comprising **Pu.1, Irf8, Id2, and Batf3** into tumor cells. The cassette reprograms glioblastoma cells toward an induced myeloid antigen-presenting-cell-like state, increasing antigen-presentation capacity and inducing innate inflammasome and pyroptosis machinery, including AIM2, caspase-1, and gasdermin D pathways. In syngeneic orthotopic glioblastoma models, mPF combined with temozolomide improved survival relative to either treatment alone, but excessive neuroinflammation and cerebral edema-like toxicity were observed in a subset of tumor-free animals. The program originated at the University of Southern California and remains preclinical.
