Overview

The mPGES-1 inhibitor agent is a small molecule therapeutic candidate currently under development by ASKA Pharmaceutical. It specifically targets microsomal prostaglandin E synthase-1 (mPGES-1), an enzyme that acts downstream of cyclooxygenase-2 (COX-2) to catalyze the conversion of prostaglandin H2 (PGH2) into prostaglandin E2 (PGE2). PGE2 is a key lipid mediator involved in the pathogenesis of pain and inflammation. By selectively inhibiting mPGES-1, this agent aims to reduce PGE2 levels while sparing other prostanoids, such as prostacyclin (PGI2), which are often suppressed by traditional non-steroidal anti-inflammatory drugs (NSAIDs) and COX-2 inhibitors. This selective mechanism is intended to provide potent anti-inflammatory and analgesic effects with a potentially improved safety profile, particularly regarding gastrointestinal and cardiovascular side effects. The drug is currently in Phase 1 clinical development.
