Overview

mPGES1 siRNA is a small interfering RNA (siRNA) designed to silence the expression of microsomal prostaglandin E synthase-1 (mPGES1), a key enzyme in the arachidonic acid cascade that converts COX-2-derived PGH2 into prostaglandin E2 (PGE2). Research presented by MD Anderson Cancer Center and the University of Pennsylvania indicates that mPGES1 is upregulated in melanoma and correlates with disease progression and reduced patient survival. By targeting mPGES1, this siRNA approach aims to reduce PGE2 production, which in preclinical models has been shown to attenuate melanoma cell survival and induce apoptosis. This strategy is being explored as a potentially safer alternative to broad COX inhibitors, which are often associated with significant systemic toxicities.
