Overview

The MPHOSPH1 peptide is a cancer vaccine candidate consisting of an HLA-A*2402-restricted epitope derived from the M-phase phosphoprotein 1 (MPHOSPH1), a kinesin-like motor protein also known as KIF20B. MPHOSPH1 is a cancer-associated antigen that is overexpressed in various malignancies, including bladder and breast cancers, while showing minimal expression in normal tissues except for the testis. The peptide is designed to be presented by the HLA-A*2402 molecule on the surface of antigen-presenting cells, thereby inducing the activation and proliferation of antigen-specific cytotoxic T lymphocytes (CTLs). These CTLs subsequently recognize and eliminate tumor cells that overexpress the MPHOSPH1 protein. In clinical settings, the peptide is typically administered via subcutaneous injection and is often emulsified with an adjuvant, such as Montanide ISA 51, to enhance the immune response. It has been investigated both as a monotherapy and as part of multi-antigen peptide cocktails targeting multiple cancer-specific molecules simultaneously.
