Overview

MPI-674 (Femathina) is a small molecule aromatase inhibitor developed by Meditrina Pharmaceuticals. It is based on the active pharmaceutical ingredient anastrozole and was repurposed for gynecologic indications, particularly for endometrial thinning prior to endometrial ablation in premenopausal women with abnormal uterine bleeding (AUB). Aromatase inhibitors like MPI-674 work by binding to and inhibiting the enzyme aromatase, which converts androgens into estrogens. By reducing estrogen production, these drugs are effective in treating estrogen-dependent conditions. While aromatase inhibitors are widely used in postmenopausal breast cancer therapy, Meditrina aimed to address unmet needs in women's health such as AUB, endometriosis, and uterine fibroids with this compound. Despite initial clinical development reaching Phase 2 trials for menstruation disturbances/AUB in the United States, further development of MPI-674 has been discontinued[1][3][4][5][9].
