MPI-676 is an orally bioavailable, small molecule inhibitor of Mucosa-associated lymphoid tissue lymphoma translocation protein 1 (MALT1), currently under development by Monopteros Therapeutics. MALT1 is a paracaspase that plays a critical role in the CARMA1-BCL10-MALT1 (CBM) complex, which mediates NF-κB signaling following antigen receptor stimulation in lymphocytes. In the context of oncology, MPI-676 is designed to selectively modulate the immune microenvironment by impairing the suppressive function of regulatory T-cells (Tregs) while preserving the activity of effector T-cells. This selective inhibition aims to overcome tumor-induced immunosuppression and enhance anti-tumor immune responses. MPI-676 is being evaluated in clinical trials for the treatment of advanced solid tumors, both as a monotherapy and in combination with immune checkpoint inhibitors.
