MPI-8 is a small molecule peptidomimetic aldehyde that acts as a potent inhibitor of the SARS-CoV-2 main protease (Mpro), also known as 3CL protease (3CLpro). Originally discovered by researchers at Texas A&M University and subsequently licensed by Sorrento Therapeutics, MPI-8 was developed as an oral therapeutic candidate for the treatment of COVID-19. The compound's mechanism of action involves forming a reversible covalent bond with the catalytic cysteine residue (Cys145) of the Mpro enzyme, which is essential for the proteolytic processing of viral polyproteins (pp1a and pp1ab) into functional units. By blocking this activity, MPI-8 prevents viral replication. Preclinical data indicated that MPI-8 possesses high antiviral potency against multiple SARS-CoV-2 variants and exhibits favorable pharmacokinetic properties, supporting its potential as an oral antiviral agent.
