**MPLA-L** is a liposomal vaccine-adjuvant formulation containing monophosphoryl lipid A, a detoxified lipopolysaccharide-derived innate immune agonist. MPLA activates Toll-like receptor 4 to enhance antigen presentation and promote cellular and humoral immune responses when co-administered with a vaccine antigen. The MPLA-L label denotes MPLA incorporated into liposomes rather than a single internationally standardized molecular entity; it has been used with HIV envelope antigens, including CN54gp140-containing investigational HIV vaccine regimens.
