MPLA liposomes is a liposomal adjuvant formulation containing monophosphoryl lipid A (MPLA), a detoxified derivative of lipopolysaccharide (LPS) that acts as a Toll-like receptor 4 (TLR4) agonist. Developed by Academisch Medisch Centrum (Amsterdam UMC) and manufactured to GMP standards by Polymun Scientific, MPLA liposomes are designed to stimulate innate immune signaling and enhance adaptive immune responses. Rather than acting as a standalone therapeutic, this formulation is used as an immune-stimulating adjuvant in combination with recombinant HIV-1 envelope glycoprotein vaccines, such as ConM SOSIP.v7 gp140, to boost humoral and cellular immunogenicity. It has been evaluated in Phase 1 clinical trials (including NCT03816137 and NCT03961438) for HIV-1 prevention in healthy, HIV-uninfected adults.
