Overview

MPN-coated engineered Lactobacillus reuteri is an experimental oral live biotherapeutic product (LBP) developed for the targeted treatment of inflammatory bowel disease (IBD), including ulcerative colitis. The therapeutic platform utilizes the probiotic bacterium *Lactobacillus reuteri* which has been genetically engineered to constitutively express and secrete interleukin-10 (IL-10), a potent anti-inflammatory cytokine. To overcome the challenges of oral delivery, the bacteria are encapsulated within a metal-phenolic network (MPN) coating, typically composed of tannic acid and ferric ions (Fe3+). This protective shell enhances bacterial survival against gastric acid and bile salts while providing intrinsic antioxidant activity by scavenging reactive oxygen species (ROS) at the site of intestinal inflammation. Upon reaching the colon, the engineered bacteria colonize the mucosa and locally deliver IL-10, which modulates the immune response by binding to IL-10 receptors on leukocytes, thereby suppressing pro-inflammatory cytokine production and promoting tissue repair.
