Overview

MPS peptide is a synthetic 25-mer peptide designed to mimic the phosphorylation site domain (PSD) of the myristoylated alanine-rich C kinase substrate (MARCKS) protein. It functions as a competitive inhibitor of MARCKS phosphorylation by protein kinase C (PKC), thereby preventing the translocation of MARCKS from the plasma membrane to the cytosol. This inhibition modulates several downstream signaling pathways, including PI3K/AKT and NF-κB, which are critical for cell proliferation, migration, and inflammatory responses. Furthermore, the MPS peptide binds to and sequesters phosphatidylinositol 4,5-bisphosphate (PIP2) at the cell membrane, further disrupting PIP2-dependent signaling cascades. Preclinical studies have demonstrated its potential in treating lung cancer by suppressing tumor growth and metastasis, in idiopathic pulmonary fibrosis by reducing fibroblast activation, and in asthma by alleviating airway hyperreactivity and inflammation.
