MPS1 is a small molecule inhibitor of the Monopolar Spindle 1 (MPS1) kinase, also known as TTK protein kinase, currently under development by Cancer Research Horizons for the treatment of multiple cancer indications. MPS1 is a dual-specificity kinase that plays a critical role in the spindle assembly checkpoint (SAC), a surveillance mechanism that ensures accurate chromosome segregation during mitosis by preventing the onset of anaphase until all chromosomes are properly attached to the spindle. By inhibiting MPS1, the drug disrupts the SAC, leading to premature mitotic exit, chromosomal instability, aneuploidy, and the induction of apoptosis (cell death) in rapidly dividing cancer cells. The asset is currently in Phase 1 clinical development to evaluate its safety, tolerability, and preliminary anti-tumor activity.
