MPT-0118 + pembrolizumab

MPT-0118 + pembrolizumab is a combination immunotherapy regimen under clinical investigation for the treatment of advanced or metastatic solid tumors. MPT-0118 is an oral small molecule inhibitor of mucosa-associated lymphoid tissue lymphoma translocation protein 1 (MALT1), designed to induce regulatory T cell (Treg) fragility and enhance anti-tumor immune responses. Pembrolizumab, marketed as Keytruda, is a humanized monoclonal antibody that targets programmed cell death protein 1 (PD-1), blocking its interaction with PD-L1 and PD-L2 to promote T-cell mediated tumor destruction. The combination aims to synergistically activate the immune system against cancer cells by both reprogramming suppressive Tregs in the tumor microenvironment and releasing inhibitory checkpoints on effector T cells. This approach is being evaluated in phase 1/1b clinical trials for multiple solid tumor types including bladder, breast, colorectal, esophageal, head and neck/oral, kidney, lung cancers, melanoma, pancreatic cancer, and prostate cancer[1][3][4].
