MQ833 is a second-generation, non-replicative recombinant modified vaccinia virus Ankara (MVA) engineered for cancer immunotherapy. Developed by Genvira Biosciences, it is designed to enhance antitumor immunity by modulating the tumor microenvironment through multiple mechanisms. The virus is modified by the deletion of three viral immunosuppressive genes: E5R (a cGAS inhibitor), E3L, and WR199, which increases its immunogenicity and activates the cGAS/STING pathway. MQ833 carries three therapeutic transgenes: membrane-bound Fms-related tyrosine kinase 3 ligand (Flt3L), OX40 ligand (OX40L), and extracellular matrix-anchored interleukin-12 (IL-12). These components work synergistically to promote myeloid cell recruitment, provide T-cell costimulation, and drive type I interferon (IFN) signaling. In preclinical studies, MQ833 has demonstrated potent antitumor effects in various solid tumor models, including ovarian cancer, and has shown superior efficacy compared to its oncolytic vaccinia counterparts.
