MQX-5906 is a topical formulation of methotrexate developed by MediQuest Therapeutics for the treatment of fingernail psoriasis. Methotrexate is a folate antimetabolite that inhibits dihydrofolate reductase (DHFR), an enzyme necessary for the synthesis of purines and thymidylate. By inhibiting DHFR, methotrexate disrupts DNA synthesis, repair, and cellular replication, which in turn reduces the hyperproliferation of keratinocytes and inflammatory activity associated with psoriasis. MQX-5906 was specifically designed to deliver methotrexate through the nail plate to treat nail bed and nail matrix psoriasis. It was evaluated in a Phase 2b dose-response study (NCT00357578) alongside other concentrations, MQX-5902 and MQX-5904, to assess efficacy and safety in improving nail appearance.
