MR-146

MR-146 is a small molecule, G-protein biased mu-opioid receptor (MOR) agonist being developed by Mebias Discovery for the treatment of moderate-to-severe pain. The drug is engineered to selectively activate the G-protein signaling pathway, which mediates analgesia, while minimizing the recruitment of beta-arrestin 2. Beta-arrestin recruitment is traditionally associated with the adverse effects of opioids, including respiratory depression, nausea, and gastrointestinal dysfunction. By optimizing this biased signaling profile, MR-146 aims to provide potent pain relief with an improved safety profile and a broader therapeutic index compared to conventional mu-opioid agonists.
