Overview

MR1-1XaCD3 is a recombinant bispecific T-cell engager (BiTE) designed for the treatment of glioblastoma multiforme (GBM). It is composed of two single-chain variable fragments (scFv): the MR1-1 scFv, which specifically targets the tumor-specific epidermal growth factor receptor variant III (EGFRvIII) mutation, and an anti-CD3 scFv that binds to the T-cell receptor complex. By cross-linking EGFRvIII-expressing tumor cells with cytotoxic T cells, MR1-1XaCD3 triggers T-cell activation and targeted tumor cell lysis. Developed by researchers at Duke University, this therapeutic candidate leverages the high specificity of the EGFRvIII mutation, which is widely expressed in GBM but absent in normal tissues, to minimize off-target toxicity.
