Overview

MR31367 is a small molecule oligopyridine derivative that acts as a direct inhibitor of the anti-apoptotic protein Mcl-1. Developed as a second-generation analog of Pyridoclax, MR31367 targets the hydrophobic binding pocket of Mcl-1, thereby displacing pro-apoptotic proteins such as Bim and Bak. This displacement triggers Bak-mediated apoptosis in cancer cells. The compound has demonstrated potent antitumor activity in chemoresistant ovarian cancer models, particularly when used in combination with Bcl-xL inhibitors like ABT-263 (navitoclax). It was developed through a collaboration involving Normandie University, INSERM, and other French research institutions.
