MRG008

MRG008 is a novel bispecific antibody-drug conjugate (ADC) designed to target both the epidermal growth factor receptor (EGFR) and trophoblast glycoprotein (5T4). Developed by Lepu Biopharma and Mabworks, the drug consists of an Fc-silenced bispecific antibody conjugated via a cleavable linker to a topoisomerase I inhibitor (Topi) payload. MRG008 is specifically engineered to address non-small cell lung cancer (NSCLC), where EGFR and 5T4 are frequently co-expressed. By targeting two distinct antigens, MRG008 aims to overcome the therapeutic limitations of single-target ADCs in patients with low or absent EGFR expression. Preclinical studies have demonstrated that MRG008 exhibits rapid internalization, potent in vitro cytotoxicity, and robust tumor growth inhibition in various NSCLC models, while maintaining a favorable pharmacokinetic profile.
