Overview

A combination **HIV-1 vaccine investigational product** consisting of two replication-incompetent recombinant adenovirus vectors: MRKAd5 HIV-1 Trigene and MRKAd6 HIV-1 Trigene. Each vector expresses three near-consensus clade B HIV-1 genes—*gag*, *pol*, and *nef*. These genes are encoded within a transgene cassette under the control of viral promoters and polyadenylation signals, designed to elicit cell-mediated immune responses. The combination vaccine aims to induce T-cell immunity against multiple HIV-1 antigens, using vectors based on adenovirus serotypes 5 (Ad5) and 6 (Ad6) to address pre-existing vector immunity in different populations. Developed and tested in phase 1 clinical trials for its safety and immunogenicity, it has not demonstrated efficacy in preventing HIV-1 infection and has not advanced to regulatory approval[2][3].
