MRKAd6 HIV-1 TRIGENE is an experimental viral vector vaccine developed by Merck for the prevention of HIV-1 infection. It utilizes a recombinant adenovirus serotype 6 (Ad6) vector, which was selected to circumvent the high levels of pre-existing immunity to the more common adenovirus serotype 5 (Ad5) found in many populations. The vector is engineered with deletions in the E1 region, which are replaced by a transgene expression cassette encoding near-consensus clade B HIV-1 Gag, Pol, and Nef proteins. The vaccine is designed to elicit robust cell-mediated immune responses, specifically CD8+ T-cell responses, against these internal HIV-1 proteins to control viral replication. Clinical evaluation focused on safety, tolerability, and immunogenicity, but development was largely impacted by the outcomes of Merck's earlier Ad5-based vaccine trials.
