Overview

MRM-ON1 is a first-in-class metabolic reprogramming molecule (MRM) developed by Zylem Biosciences that targets Scavenger Receptor Class B Type 1 (SR-B1). SR-B1 is a cell surface receptor responsible for the selective uptake of cholesteryl esters from high-density lipoprotein (HDL). Many cancer cells, including those in ovarian, prostate, and pancreatic tumors, as well as hematological malignancies like acute myeloid leukemia (AML) and diffuse large B-cell lymphoma (DLBCL), overexpress SR-B1 to satisfy their increased demand for cholesterol, which is vital for membrane biogenesis and proliferative signaling. MRM-ON1 acts as an SR-B1 antagonist, effectively starving cancer cells of cholesterol and inducing cell death. The program is currently in the IND-enabling phase for several solid tumor indications and in preclinical development for various blood cancers.
