mRNA-0217 (also referred to as mRNA-0217/S001) is an investigational personalized messenger RNA (mRNA) tumor vaccine developed through a collaboration between Ruijin Hospital (Shanghai Jiao Tong University School of Medicine) and Shanghai deepGene Biotechnology (deepGeneAI). The vaccine utilizes an artificial intelligence-driven platform to identify up to 20 unique tumor-specific neoantigens from each patient's genomic profile through next-generation sequencing. These patient-specific neoantigens are encoded into mRNA and delivered via lipid nanoparticles (LNPs). Once administered, the mRNA is translated into proteins that are presented to the immune system, primarily aiming to stimulate robust neoantigen-specific CD4+ and CD8+ T-lymphocyte responses to recognize and destroy tumor cells. mRNA-0217 is typically investigated as an investigator-initiated trial (IIT) therapy for advanced solid tumors and pancreatic cancer, often in combination with the immune checkpoint inhibitor pembrolizumab to enhance therapeutic efficacy.
