mRNA-1273.351 is an investigational, variant-specific mRNA-based vaccine developed by Moderna to target the SARS-CoV-2 B.1.351 (Beta) variant, first identified in South Africa[2][5][6]. It encodes a modified version of the spike (S) glycoprotein of the B.1.351 lineage of SARS-CoV-2 and is delivered using lipid nanoparticle technology[3]. The vaccine is designed as a booster or primary immunization to enhance immune responses against variants of concern, particularly those with mutations that may reduce neutralization by antibodies elicited by earlier vaccines[5][8]. Clinical studies have shown that a booster dose of mRNA-1273.351 increases neutralizing antibody titers against both the original virus and variants such as B.1.351 and P.1, with higher titers observed for B.1.351 compared to boosters with the original mRNA-1273 formulation[5][8]. The mechanism of action involves induction of an immune response targeting the coronavirus spike glycoprotein, thereby preventing infection.
