Overview

mRNA-1273.529 is an investigational, Omicron-specific (B.1.1.529) COVID-19 vaccine candidate developed by Moderna as a booster to enhance immunity against the SARS-CoV-2 Omicron variant. It is based on messenger RNA (mRNA) technology and encodes the spike protein of the Omicron BA.1 subvariant, aiming to induce neutralizing antibodies specifically targeting this variant. Preclinical studies in mice demonstrated that a primary vaccination series with mRNA-1273.529 potently neutralized B.1.1.529 but showed limited inhibition of historical or other SARS-CoV-2 variants; boosting with either mRNA-1273 or mRNA-1273.529 increased serum neutralizing titers and protection against B.1.1.529 infection. The vaccine has been evaluated in Phase 2/3 clinical trials as both a primary series and booster dose in adults previously vaccinated with earlier versions of Moderna's COVID vaccines.
