mRNA-1283 (mNEXSPIKE) is a next-generation, investigational mRNA-based COVID-19 vaccine developed by Moderna. Unlike the first-generation vaccine (mRNA-1273/Spikevax), which encodes the full-length SARS-CoV-2 spike protein, mRNA-1283 encodes only the N-terminal domain (NTD) and receptor-binding domain (RBD) of the spike protein. These domains are linked to a transmembrane domain of the influenza hemagglutinin (HATM) to facilitate cell surface anchoring and antigen presentation. This refined design allows for a significantly lower dose (10 µg) compared to the original vaccine and provides enhanced stability, enabling storage at standard refrigerator temperatures (2-8°C). mRNA-1283 is utilized as a standalone vaccine and as the COVID-19 component of Moderna's combination respiratory vaccines, such as mRNA-1083 (mCOMBRIAX). In clinical trials like NCT06864143, specific batches designated as "Investigational COVID-19 Vaccine Lot A" are used as active comparators to evaluate the immunogenicity and safety of combination vaccine candidates in various adult populations.
