mRNA-1283.251 is a next-generation mRNA vaccine formulation developed by Moderna for the prevention of COVID-19. It is a variant-specific version of the mRNA-1283 (mNEXSPIKE) platform, which utilizes lipid nanoparticle (LNP) technology to deliver mRNA encoding a chimeric SARS-CoV-2 spike protein. This chimeric antigen, designated NTD-RBD-HATM, consists of the N-terminal domain (NTD) and receptor-binding domain (RBD) of the SARS-CoV-2 spike protein anchored to an influenza hemagglutinin transmembrane domain. A significant advantage of the mRNA-1283 platform is its stability at standard refrigerated temperatures (2-5 °C), which simplifies distribution compared to first-generation mRNA vaccines requiring ultra-low temperature storage. mRNA-1283.251 is specifically designed to target circulating variants and is being evaluated in Phase 3b/4 clinical trials to assess its immunogenicity and safety in older adults and high-risk populations.
