mRNA-1325 is an experimental mRNA-based vaccine developed by Moderna for the prevention of Zika virus infection. It was the first Zika vaccine candidate to enter clinical trials using Moderna's messenger RNA technology, which involves encoding viral antigens—specifically the pre-membrane (prM) and envelope (E) proteins of the Zika virus—within a lipid nanoparticle (LNP) delivery system. The development of mRNA-1325 was accelerated by significant non-dilutive funding from the Biomedical Advanced Research and Development Authority (BARDA) and the Defense Advanced Research Projects Agency (DARPA) in response to the 2015-2016 Zika outbreak. Although it successfully completed a Phase 1 dose-ranging study (NCT02964390) in healthy adults, Moderna subsequently prioritized a second-generation Zika vaccine candidate, mRNA-1893, which demonstrated a more robust profile.
