mRNA-1468 is an investigational mRNA-based vaccine developed by Moderna for the prevention of herpes zoster (shingles), which is caused by reactivation of latent varicella-zoster virus (VZV). The vaccine encodes the VZV glycoprotein E (gE) antigen, aiming to stimulate robust immune responses, including both CD4+ and CD8+ T cell activation. Clinical studies have shown that two doses of mRNA-1468 elicit strong antigen-specific T cell responses comparable or superior to those induced by Shingrix, a currently approved shingles vaccine. The reactogenicity profile of mRNA-1468 is similar to Shingrix. The primary indication under investigation is herpes zoster in adults aged 50 years and older[1][2][3][4][5].
