mRNA-1647 is an investigational messenger RNA (mRNA)-based vaccine developed to prevent infection by cytomegalovirus (CMV), the most common infectious cause of birth defects in the United States. The vaccine consists of six separate mRNAs encoding two key CMV antigens—five encode subunits of the membrane-bound pentamer complex, and one encodes glycoprotein B (gB). Both antigens are essential for viral entry into epithelial cells, a critical step in CMV infection. Delivered via lipid nanoparticle formulation and administered intramuscularly, mRNA-1647 instructs host cells to produce these viral proteins, thereby eliciting an immune response that includes neutralizing antibodies against both epithelial cell and fibroblast infection. The vaccine is currently being evaluated in Phase 3 clinical trials for prevention of primary CMV infection in women aged 16–40 years, with additional studies ongoing in adolescents and transplant patients[2][3][5][6][8].
