mRNA-2416 is an investigational cancer immunotherapy developed by Moderna. It consists of a synthetic messenger RNA (mRNA) encoding the human co-stimulatory protein OX40 Ligand (OX40L; also known as tumor necrosis factor ligand superfamily member 4, TNFSF4), encapsulated within a lipid nanoparticle for intratumoral administration. The drug is designed to stimulate immune responses against tumors by increasing expression of OX40L in the tumor microenvironment, thereby enhancing T-cell activation and antitumor immunity. Clinical studies have shown that intratumoral injection of mRNA-2416 increases local OX40L protein levels and pro-inflammatory activity in solid tumors. Development was primarily focused on ovarian cancer, lymphoma, and other solid tumors; however, clinical trials were discontinued after efficacy endpoints were not met[1][2][5].
