Overview

**mRNA-2752** is an investigational intratumoral lipid nanoparticle mRNA immunotherapy developed by Moderna. It co-encapsulates mRNAs encoding human OX40 ligand, interleukin-23, and interleukin-36 gamma, enabling local production of these immunostimulatory proteins within the tumor microenvironment. The expressed factors are intended to activate OX40 costimulation and proinflammatory cytokine signaling, enhance antitumor immune-cell activity, and convert immunologically inactive tumors toward an inflamed phenotype. It has been evaluated as monotherapy and with immune checkpoint inhibitors including durvalumab and pembrolizumab in advanced malignancies and high-risk ductal carcinoma in situ.
