**mRNA-2752 + durvalumab** is a combination immunotherapy currently under clinical investigation for advanced solid tumors and lymphoma. mRNA-2752 is a novel, intratumorally injected, lipid nanoparticle-encapsulated mRNA-based therapy encoding three immune-modulatory agents: OX40 ligand (OX40L), interleukin-23 (IL-23), and interleukin-36 gamma (IL-36γ). Its primary function is to enhance local and systemic anti-tumor immune responses by stimulating T-cell activation (through OX40L) and augmenting pro-inflammatory cytokine signaling (via IL-23 and IL-36γ). Durvalumab is a monoclonal antibody targeting programmed death-ligand 1 (PD-L1), acting as an immune checkpoint inhibitor to prevent PD-L1-mediated immune suppression. The combination is designed to simultaneously activate immunity in the tumor microenvironment (by mRNA-2752) and release systemic immunosuppression (by durvalumab) to enhance anti-tumor activity. This combination is primarily being studied in patients with advanced/metastatic solid tumors or lymphoma who have progressed after standard treatments[1][3][7].
