mRNA-6213 is an investigational messenger RNA (mRNA) therapy designed to encode the cyclic GMP-AMP synthase (cGAS) protein. Developed by Moderna, it is typically encapsulated in lipid nanoparticles (LNPs) for delivery. Upon entering the cytoplasm of target cells, the mRNA is translated into functional cGAS enzymes. cGAS acts as a pattern recognition receptor that detects cytosolic DNA and catalyzes the synthesis of cyclic GMP-AMP (cGAMP). cGAMP then functions as a second messenger that binds to and activates the Stimulator of Interferon Genes (STING) protein. Activation of the cGAS-STING pathway triggers the production of Type I interferons and other pro-inflammatory cytokines, thereby stimulating a potent innate and adaptive immune response against tumors. This approach aims to overcome the limitations of direct STING agonists, such as poor pharmacokinetics and narrow therapeutic windows, by leveraging the cell's own machinery to produce the upstream activator.
