The mRNA COVID-19 vaccines, specifically BNT162b2 (Pfizer-BioNTech) and mRNA-1273 (Moderna), are lipid nanoparticle-encapsulated mRNA therapies designed to prevent COVID-19 by inducing an immune response against the SARS-CoV-2 spike protein. In the context of the COVER (COVID-19 VaccinE Response in Rheumatology patients) trial, these vaccines were evaluated as booster doses in patients with autoimmune rheumatic diseases such as rheumatoid arthritis, psoriatic arthritis, and axial spondyloarthritis. The study, led by Dr. Jeffrey Curtis at the University of Alabama at Birmingham, specifically investigated whether a temporary two-week interruption of immunomodulatory treatments (e.g., TNF inhibitors, JAK inhibitors, or abatacept) at the time of vaccination could improve the magnitude of the antibody response without causing significant disease flares.
