mRNA-DC-CTL cells are a form of personalized cellular immunotherapy in which autologous dendritic cells (DCs) are pulsed or electroporated with messenger RNA (mRNA) encoding tumor-associated antigens. These antigen-loaded DCs are then used to stimulate and expand autologous cytotoxic T lymphocytes (CTLs), which can specifically recognize and kill tumor cells expressing the encoded antigens. The process involves isolating mononuclear cells from the patient, generating DCs ex vivo, loading them with relevant tumor antigen mRNAs, co-culturing them with autologous T-cells to generate CTLs, and then infusing these activated CTLs back into the patient. This approach aims to induce a robust anti-tumor immune response by harnessing both antigen presentation by DCs and direct tumor cell killing by CTLs. It is being investigated primarily for cancer indications such as glioma, melanoma, non-small cell lung cancer (NSCLC), and other solid tumors[2][3][5].
